Brno Airport (BRQ) to the city
Brno is two zones, so the airport bus needs the 60 minute ticket at 25 CZK; the 15 minute one at 20 CZK will not get you into town.
Within Brno you are always in two zones, 100 and 101. A two zone ticket costs 20 CZK for 15 minutes or 25 CZK for 60 minutes.
The 15 minute ticket does not reach the centre from the airport. Buy the 60 minute one; the 5 CZK saving is not worth the fine.
A 24 hour ticket for zones 100 and 101 is 90 CZK, 45 CZK reduced, which pays off from the fourth journey.
Line 21 runs Hlavni nadrazi to Letiste Turany, the main station to the airport.
Line 77 runs from Uzka, the stop opposite the Vankovka shopping gallery.
Night line N89 covers the airport after the day lines stop, from Hlavni nadrazi, and its stop there is on the tram tracks at track 4. That is easy to miss at night.
Buy before boarding. You cannot buy on board and you cannot reserve a seat.
A rail ticket between Brno and Vienna Airport carries an exemption in zones 100 and 101, so check what your through ticket already covers.

Everything inside Brno is a two zone journey, so the choice is only about time. The airport run does not fit into 15 minutes, which makes 25 CZK the fare for practical purposes.
Five crowns is the whole difference. Buying the short ticket to save it is how visitors end up paying a penalty fare instead.
The 24 hour ticket at 90 CZK is worth it on a day of moving around, not for a single transfer in.

21 is the one for the railway station.
77 starts at Uzka, opposite the Vankovka gallery, which is closer to the centre for some hotels.
N89 is the night service, and its main station stop is on the tram tracks at track 4 rather than at the bus stands.
Tickets
The airport is explicit that you must buy the ticket before boarding and that neither the journey nor a seat can be reserved.
Buy from a machine, a kiosk or the app before you get on, and validate as you board.
Through rail tickets between Brno and Vienna Airport carry an exemption in zones 100 and 101, which is worth checking if you are using the Vienna alternative.
